Properties of particles in sap
TIP:
55-62.5-70 °C. LIV: 0.2-1.533-2 days. DEP: log10 minus 2 (1/2), or 3 (2/2).
Infectivity of sap decreased by treatment with di-ethyl ether (1/2), or not
changed by treatment with di-ethyl ether (1/2). Leaf sap contains few virions
(1/1).
Particle morphology
Virions isometric (1/2), or unusually
shaped (1/2); not enveloped (1/1); 50-70-90 nm in diameter; rounded in profile
(1/1).
Physical properties
One sedimenting component in purified
preparations (2/2); sedimentation coefficient 270 S. Density 1.15 g
cm-3 in CsCl.
Biochemical properties
Genome consists of RNA (4/4);
single-stranded (4/4); linear (3/3). Total genome size 4.203-5.284-6.9 kb.
Genome unipartite (3/3); largest (or only) genome part 2.05-3.667-4.75 kb. Base
composition 27 % G; 25 % A; 25.5 % C; 22.4 % U. Infectivity retained when
deproteinised with proteases (1/1); retained when deproteinised with phenol or
detergent (1/1). Poly A region absent (1/1). Additional factor not required for
infectivity (2/2). Genome has no tRNA-like activity (1/1).
Features of the genome
Non-genomic nucleic acid
found in the virions (2/2); is subgenomic mRNA (1/2), or satellite RNA
(1/2). Sub-genomic mRNA found in infected cells (2/2). 2 virus
specified dsRNA species found in infected cells. Size of largest virus specified
dsRNA 4.2 kbp; 2nd largest 1.5 kbp.
Features of proteins
Virion protein(s) none (1/1).
Replication
Genome replicates in cytoplasm (1/1).
Replication depends on co-infection with a helper virus (1/2), or does not
depend on a helper virus (1/2). Acts as helper for a satellite virus (1/1).
Cytopathology
Inclusions present in infected cells (1/1);
are membranous bodies (1/1); they do not contain virions (1/1).
