Physical and
biochemical properties
Properties of particles in sap
TIP:
80-87.5-95 °C. LIV: 7-46.83-63 days. DEP: log10 minus 4 (1), or 7 (1), or
9-10 (1). Leaf sap contains few virions (1/2), or contains many virions (1/2).
Particle morphology
Virions isometric; not enveloped
(2/2); 26-28.66-30 nm in diameter; rounded in profile (1/2), or angular in
profile (1/2); without a conspicuous capsomere arrangement.
Physical properties
One sedimenting component in purified
preparations; sedimentation coefficient 99-118.6-136 S. Density
1.333-1.367-1.399 g cm-3 in CsCl. Isoelectric point pH 4.5.
A260/A280 ratio 1.61.
Biochemical properties
Virions contain 18-18.5-19 %
nucleic acid; 72-76.5-81 % protein; 0 % lipid.
Genome consists of RNA; single-stranded; linear (1/1). Total genome size
3.759-3.981-4.185 kb. Genome unipartite (2/2); largest (or only) genome part
3.759-3.879-4 kb. Base composition 24 % G; 27.5 % A; 22.5 % C; 25.5 % U. 5´
terminus of RNA has an unusual type (1/1). Infectivity retained when
deproteinised with phenol or detergent (1/1). Poly A region absent (1/1).
Features of the genome
Non-genomic nucleic acid
not found in the virions (1/1). Sub-genomic mRNA found in infected
cells (1/1).
Features of proteins
Virion protein(s) one;
Mr 27000-30830-35500.
Replication
Replication does not depend on a helper virus
(1/1).
Cytopathology
Virions found in cytoplasm (2/2).
Inclusions present in infected cells (2/2); are crystals in the cytoplasm (2/2),
or membranous bodies (1/2), or unusual in shape (1/2); they contain virions
(1/2), or they do not contain virions (1/2).
Taxonomy and relationships
