Purification method
Cockbain: grind
frozen tissue in 0.5M phosphate buffer, pH 8.0 containing 0.01M EDTA and 0.4%
2-mercaptoethanol with chloroform/butanol (1 g leaf: 2 ml buffer: 0.5 ml
chloroform: 0.5 ml N-butanol) followed by differential centrifugation.
Particle morphology
Virions isometric; not enveloped; 30
nm in diameter; angular in profile; without a conspicuous capsomere arrangement.
Physical properties
Density 1.38 and 1.39 g cm-3 in
CsCl.

Virus(es) with serologically unrelated virions
Bean leaf roll, broad bean mottle, broad bean true mosaic, broad bean
wilt, carnation cryptic, crimson clover latent, pea enation mosaic and southern
bean mosaic viruses.
