Properties of particles in sap
Leaf sap
contains many virions. Electron microscopy: negatively-stained virions from cut
leaf edges, better than sap extracts.

Particle morphology
Virions filamentous; not enveloped;
usually flexuous; with a clear modal length; of 749 nm; 15 nm wide. Axial canal
obscure. Basic helix obscure.
Physical properties
One sedimenting component in purified
preparations. A260/A280 ratio 1.038.
Replication
Replication does not depend on a helper
virus.
Cytopathology
Virions found in leaves, mesophyll and
meristems; in cytoplasm. Inclusions present in infected cells; are unusual in
shape; stacked plates (laminated aggregates) thought to be parts of cylindrical
or pinwheel inclusions; they do not contain virions.
